چکیده

Background:
DNA-alkyltransferases repair mutagenic and carcinogenic alkylation DNA lesions. Results:
A thermophilic archaeal DNAalkyltransferase is degraded after alkylation in vivo. A novel assay is applied to study its activity in vitro. Conclusion:
The archaeal DNA-alkyltransferase shows structure, activity and in vivo regulation similar to its human homolog. Significance:
The function and regulation of DNA-alkyltransferases might be conserved from archaea to humans.

A bifunctional DNA repair protein from Ferroplasma acidarmanus exhibits O6-alkylguanine-DNA alkyltransferase and endonuclease V activities.

A recently discovered DNA repair protein of 303 aa from the archaeal organism Ferroplasma acidarmanus was studied. This protein (AGTendoV) consists of a fusion of the C-terminal active site domain of O(6)-alkylguanine-DNA alkyltransferase (AGT) with an endonuclease V domain. The AGTendoV recombinant protein expressed in Escherichia coli and purified to homogeneity repaired O(6)-methylguanine le...

Structure-function relationships governing activity and stability of a DNA alkylation damage repair thermostable protein

Alkylated DNA-protein alkyltransferases repair alkylated DNA bases, which are among the most common DNA lesions, and are evolutionary conserved, from prokaryotes to higher eukaryotes. The human ortholog, hAGT, is involved in resistance to alkylating chemotherapy drugs. We report here on the alkylated DNA-protein alkyltransferase, SsOGT, from an archaeal species living at high temperature, a con...

High level, regulated expression of the chimeric P-enolpyruvate carboxykinase (GTP)-bacterial O6-alkylguanine-DNA alkyltransferase (ada) gene in transgenic mice.

Transgenic animals expressing genes capable of repairing DNA may be a valuable tool to study the effect of DNA-damaging agents on tissue-specific carcinogenesis. For this reason, we constructed a chimeric gene consisting of the promoter-regulatory region of the phosphoenolpyruvate carboxykinase (GTP) (EC 4.1.1.32) (PEPCK) gene linked to the Escherichia coli ada gene coding for O6-alkylguanine-D...

Regeneration of 06-Alkylguanine-DNA Alkyltransferase in Human Lymphocytes after Nitrosourea Exposure1

Mitogen-stimulated human lymphocytes have an increased capacity to repair many forms of DNA damage caused by IV, ionizing radiation, and chemical carcinogens. Human lymphocytes rely on a particular DNA repair protein, O'-alkylguanine-DNA alkyltransferase (alkyltransferase) to repair efficiently O6-alkylguanine, an important mutagenic adduct formed by nitrosoureas and other /V-nitroso compounds....

Thermostable archaeal O6-alkylguanine-DNA alkyltransferases.

Archaea represent some of the most ancient organisms on earth, and they have relatively uncharacterized DNA repair processes. We now show, using an in vitro assay, that extracts of two Crenarchaeota (Sulfolobus acidocaldarius and Pyrobaculum islandicum) and two Euryarchaeota (Pyrococcus furiosus and Thermococcus litoralis) contain the DNA repair protein O6-alkylguanine-DNA alkyltransferase (ATa...
